Putative tripeptidyl peptidase in renal brush border
To the Editor: We wish to draw readers’ attention to the need for caution in interpreting assays of peptidases using fluorogenic substrates such as Gly-Pro-Met-2 naphthylamide (NNap). The release of the fluorophore, 2naphthylamine, may indicate the action of a tripeptidyl peptidase releasing the tripeptide, Gly-Pro-Met, but it may also result from the sequential attack of other exopeptidases, f...
متن کاملGlycosaminoglycans from renal brush border membranes of rabbits.
Brush border membranes were prepared from the rabbit kidney cortex by sucrose density gradient centrifugation and digested with pronase. A glycosaminoglycan fraction was isolated from the digest by ion-exchange chromatography. Electrophoretogram on cellulose acetate membrane (Separax) of the glycosaminoglycan fraction indicated the presence of two components, which were identified as chondroiti...
متن کاملTaurine transport in renal brush-border-membrane vesicles.
Taurine transport in isolated brush-border-membrane vesicles from rat kidney is concentrative and it is driven by the Na+ gradient and transmembrane potential difference; binding is not a significant component of net uptake. The Na+-dependent component of net uptake is saturable with an apparent Km of 17 microM. The taurine-transport mechanism is selective for beta-amino compounds.
متن کاملKinetics of sodium succinate cotransport across renal brush-border membranes.
The kinetics of Na/succinate cotransport across renal brush-borders was studied using membrane vesicles. Initial rates of succinate uptake (Js) were estimated from 1-s uptakes measured under voltage-clamped conditions. Lowering the external (cis) sodium concentration reduced the succinate Kt without affecting the Jsmax. Increasing the intravesicular (trans) sodium concentration reduced both Jsm...
متن کاملNHE3: a Na+/H+ exchanger isoform of renal brush border.
Na+/H+ exchangers in the brush-border (luminal, apical) membrane of renal proximal tubules are responsible for active, transcellular reabsorption of NaHCO3 and NaCl. Although well characterized kinetically, the protein that mediates Na+/H+ exchange in the renal brush border has not been identified. Several Na+/H+ exchanger genes, including NHE1, NHE2, NHE3, and NHE4, are expressed in the kidney...
